Bring up task Manager - see what is running. Run msconfig see what fires up on windows starts up. If you are unsure about a particular process name - google it.

Have you run a virus scan and spyware clean up recently ?

I think there are numerous shareware / freeware / trial ware programs that offer a more complete analysis of "what is my machine doing". (again google)

Lots of programs access the hard drive intermittenly good ones as well as not so good ones. Tune XP is a pretty safe program (I use it) so after a spyware / virus scan and checking runing processes you should feel better :D

Actually Tune XP tweaks should minimize access. Like removing the time stamping thing for NTFS.

Windows accesses the drive when your system is idling to index it. Try switching off that, from My Computer > right click drive > properties > down on the bottom

I had nothing but problems after installing TuneXP so I removed it, all is well.

Depending on what aspect of the OS that you wish to "tune", have a look at the XP Power Toys package, specifically the TweakUI application. It will alow you to change certain aspects of the XP user interface, e.g. start menu, logon screen, etc. You can get it here.
The power toys application is not officially supported by Microsoft, but it was written by the same developers who worked on XP, so it is probably safer & more reliable than a 3rd party tuning application.
