I'm using Vista Business (64 bit) and have a fingerprint reader, could I make it so people would need a fingerprint to access certain files?

Yes, it'll run via your fingerprint readers software.
Find the software that goes with your fingerprint reader - either it will inlcude encryption or you should be able to get compatible software.
However: 64 Bit makes life difficult...

(My Sony SZ 32 Bit Vista can do that)
