Ive read about this happening with other laptops. I took my laptop (W500) apart to upgrade the ram and when all the screws went back, struggling with the last one above the product key, my speakers didn't work. They said they may have been on too tight or something (screws). So I removed the screw about the product key and they work.....

Do I need this screw or is it ok to be 4/5?
What causes the speakers to not work out of curiosity?

You probably have the screws mixed up. Somewhere there's a screw that's a little shorter than the hole it's in.

I'm inclined to agree with cereal killer, as you only need to remove four screws to be able to swap ram in a W500. If you took out five screws, then chances are that 5th one got put back in the wrong place.
