The drive is fastest in the beginning and slowest at the end. My 500GB Toshiba varies from 75MB/s in the beginning to 35MB/s at the end with a mean speed of 55MB/s. My drive is partitioned as follows:

    0-100GB Windows 7 + essential apps
100-282GB Games and non essential apps
    282-465GB Backups, Installfiles, Other stuff

A 320GB could be partitioned as:

    0-50GB Windows Vista/7 + essential apps
    50-149GB Games other apps
    149-298GB Backups, Installfiles, Other stuff

You would probably partition it another way to suit your needs but for the fastest boot time keep the os as close to the beginning of the drive as possible. Also make sure you use the latest drivers. My boot time is about 35s to login screen and 42-45s to desktop (depending on how fast i enter the password).

If you're asking this many questions, chance are nothing less than a clean install will work for you. Start from scratch, and custom install everything exactly the way you want it.

System feels snapier and when i did a HD tune benchmark and look at it... i was like Wow... the Burst speed was just absurd! I did 3 partitions of 60GB for boot and 100GB for games and 138GB for data...
