I'm looking at several ASUS laptops and some have Windows 7 Home Premium in the package while others have Windows Vista w/ Windows 7 Home Premium Upgrade Coupon. If I get the latter should I upgrade to Windows 7 Home Premium for $119 or use the coupon and do a fresh Windows 7 install right after I receive the computer? Is it a hassle to do the upgrade?

You should get the Windows 7 upgrade for free.

And it is easy to do. Either do the super-easy upgrade, or back your stuff up and do a clean install.
