If I partition an external HD that's formatted with FAT32 with a Mac partition, will I lose any data I have on it already?

    It's likely that you would.

What do you mean? if you mantain the original FAT32 partition and add another Mac partition at the end, then it is possible that you won't loose any data - providing data is not fragmented and not at the place the new partition will take, obviously. But there is always a risk, a backup is highly recommended in re-partitioning.
