I applied arctic silver 5 to my GPU/CPU and let it sit for a day, but then took the heat sink off again the next day to check for a voltage regulator. I then just placed the heat sink right back on. Is there some rule like if you take off the heat sink you have to reapply thermal grease?

Yes, and you just said the rule:
    :D

Otherwise there will be little air bubbles between the die and the heatsink which will nullify the effect of the thermal paste.
